WHY?
Fluorescence imaging can suffer from non-specific signal.
Non-specific tracer uptake, on-target uptake but off site or even reflected light can cause non-specific signal in your fluorescence imaging which reduces the target to background contrast or can even lead to false positives!
SOLUTION: Image the time dimension
The time dimension of fluorescence adds a new layer of information which can show true from false signal
The specific signal has a specific time time-domain color and can be separated from the false signals.
HOW?
A powerful laser illuminates the field of view with fast pulses of light. Pulsed illumination concentrates the light by a factor ~1000 and makes it stand out from background sources like room lights.
The pulsed excitation of fluorescence results in the emission of fluorescence decays instead of continuous fluorescence.
The tauCAM fast-gated NIR camera can gate out the individual decays, suppressing the unwated background light.
By changing the position of the tauCAM gate within the fluorescence decay, the fluorescence lifetime can be detected. Either measure the fluorescence in two positions A and B and get a rapid estimation of the lifetime by taking the ratio of the signal in position A versus B or sweep the gate through the decay with small steps and fit the full decay to your model or convert it to the frequency domain for a phasor analysis.
Taurion taupologyTM software will map the detected lifetimes to a new color scale offering you a time-domain color reveiling information that was not visible on the conventional intensity scale.
